چکیده

Hyaluronan (HA) is a high molecular weight linear glycosaminoglycan and abundant in the skin extracellular matrix. Cutaneous HA first depolymerized to intermediate sizes then further fragmented regional lymph nodes. High anti-inflammatory anti-angiogenic, whereas pro-inflammatory pro-angiogenic. We have reported that HYBID (HA-Binding protein Involved Depolymerization), also known as KIAA1199/CEMIP, responsible for step of depolymerization, but it was recently shown mouse TMEM2 (mTMEM2) with structural similarity HYBID, membrane-bound hyaluronidase. previously showed essential depolymerization normal human dermal fibroblasts (NHDF), knockdown (hTMEM2) enhanced depolymerization. The purpose this study elucidate degradation activity functions hTMEM2. expression plasmids were transfected into HEK293T cells, each confirmed. hHYBID mTMEM2 degraded HA, hTMEM2 did not, indicating does not function Then, we focused on amino acid residues are conserved HA-degrading hHYBID, mHYBID, substituted Simultaneous replacement corresponding (H248N A303F) GG domain abolished activity. In addition, stimulation NHDF by IL-1β increased mRNA subsequently decreased along an increase HAS2-dependent production, which reversed knockdown. Taken together, these results indicate hyaluronidase regulator metabolism.

Mouse testicular hyaluronidase-like proteins SPAM1 and HYAL5 but not HYALP1 degrade hyaluronan.

Besides SPAM1 (sperm adhesion molecule 1; formerly named PH-20), further hyaluronidase-like proteins, HYAL5 (hyaluronoglucosaminidase 5) and HYALP1 (hyaluronoglucosaminidase pseudogene 1) are also expressed in murine testicular tissue. As they share a high degree of sequence similarity with known hyaluronidases, all three polypeptides could potentially exhibit hyaluronidase activity, a function...

CD93 is Selectively Expressed on Human Myeloma Cells but Not on B Lymphocytes

Background: CD93 has originally been known as a C1q receptor, and many studies have demonstrated that CD93 is expressed on hematopoietic stem cells, B cell progenitors, myeloid and monocytic cells. Moreover, CD93 is shown to be expressed on long-lived plasma cells, and CD93 deficient-mice display an impairment in plasma cell development. Objective: To investiga...

Hyaluronan is not just a goo!

Long, long ago, when hyaluronan (HA; also termed hyaluronic acid or hyaluronate) was just a goo, a trickle of papers emerged suggesting that it might also play an important role during embryonic development, possibly creating hydrated pathways that facilitate cell movement (reviewed in ref. 1). Among these early publications were studies by Roger Markwald’s group indicating that endothelial cel...

HABP2 is a Novel Regulator of Hyaluronan-Mediated Human Lung Cancer Progression

BACKGROUND Lung cancer is a devastating disease with limited treatment options. Many lung cancers have changes in their microenvironment including upregulation of the extracellular matrix glycosaminoglycan, hyaluronan (HA), which we have previously demonstrated can regulate the activity of the extracellular serine protease, hyaluronan binding protein 2 (HABP2). This study examined the functiona...

Hyaluronan, a Crucial Regulator of Inflammation

Hyaluronan (HA), a major component of the extracellular matrix (ECM), plays a key role in regulating inflammation. Inflammation is associated with accumulation and turnover of HA polymers by multiple cell types. Increasingly through the years, HA has become recognized as an active participant in inflammatory, angiogenic, fibrotic, and cancer promoting processes. HA and its binding proteins regu...
